6.Optimization of Cronobacter dublinensis subsp. dublinensis DES187(T) isolated from root nodules of Soybean for exopolysaccharide production

In present study conditions were optimized by one-factor-at a time method to enhance EPS production by Cronobacter dublinensis subsp. dublinensis DES187(T) isolated from root nodules of Soybean (Glycin max). Effect of different parameters were examined on EPS production and maximum EPS production was observed with 2% D-Glucose as carbon source (750μg/ml), 0.2% beef extract as nitrogen source (770μg/ml) and with 300μg/ml of MgSo4.7H2O as metal ion (820μg/ml) after 66 hours of incubation period (880μg/ml). EPS was harvested with ethanol and estimated by phenol sulphuric acid assay. With all this optimized conditions Cronobacter dublinensis subsp. dublinensis DES187(T) produced 4.0 g/l exopolysaccharide as per dry weight and 920µg/ml of exopolysaccharide estimated by phenol sulphuric acid assay.
